Abstract
CHACON MARCHECO, Edilberto et al. Molecular identification of Toxocara canis in canines in Salcedo canton, Ecuador. RCTU [online]. 2022, vol.9, n.1, pp.66-74. ISSN 1390-7697. https://doi.org/10.26423/rctu.v9i1.679.
Nematodes constitute an important public health problem, therefore, the identification of Toxocara canis from canine isolates in the city of Salcedo, Cotopaxi province, Ecuador, by sequencing the second internal transcribed spacer region (ITS-2) of ribosomal DNA (rDNA), constitutes an effective tool addressed in the present study. Adult parasites of the genus T. canis were sampled. The phenol-chloroform method allowed DNA extraction. PCR amplification was performed using specific primers and the product was sequenced by capillary electrophoresis. A total of 383 bp of the ITS-2 ribosomal DNA gene were sequenced. The identity of each isolate was confirmed using the NCBI BLAST algorithm. Phylogenetic trees were constructed using the MEGA X program. The 20 canine isolates were identified as Toxocara canis, when compared with sequences previously deposited in GenBank. The phylogenetic tree confirmed the identity of the studied isolates as belonging to the genus Toxocara, particularly T. canis.
